Voordat Clarke zich specialiseerde in de basgitaar, bespeelde hij de accordeon, de viool en de cello. Clarke is briljant op zowel de akoestische als elektrische bas. Hoewel hij naast de jazz zich ook in veel andere muziekkringen (r&b, pop en klassiek) heeft bewogen, heeft Clarke toch kans gezien met de beste jazzlegendes op aarde te spelen.

Vanaf midden jaren zeventig funkte hij bij George Duke in de band, met wie hij diverse succesplaten maakte. Clarke is een complete musicus, die naast zijn gloedvolle optredens ook veelvuldig componeert. Een grote liefde is het schrijven en uitvoeren van grote orkestrale werken voor films en series (bekendste: Boys N the Hood, What's Love Got to Do With It en Romeo Must Die).

Zijn naam werd groot toen hij in Chick Corea's 'Return to Forever' terecht kwam, waar zijn basgeluid een belangrijke motor was in het rockgeori‘nteerde fusion kwartet.

In 1980 bracht hij het legendarische album 'Rocks, Pebbles & Sand' uit, dat spettert van de funk-gitaarrifs en solo's.
